چکیده

Early stud& with pyrimidines administered t.o animals established the fact that, these compounds arc metabolized (3, 4). Present concepts of the pathways of pyrimidine metabolism stem largely from the work of Cerecedo (4), in which he measured the changes in the urinary excretion of urea after feeding pyrimidines and related subst.ances. On the basis of these studies, he proposed an init.ial oxidation at carbon 5, which in the case of uracil yields isobarbituric acid and with t.hymine results in thymine glycol. It was furt.her proposed that oxalic acid, formic acid, and urea are ult.imately obtained from uracil and t,hat acetol, carbon dioxide, and urea result from thymine. However, the indirect nature of the evidence leaves this scheme open t’o question. Our approach to this problem has been first to obtain, through enrichment culture, bact.erial strains which rapidly metabolize pyrimidines and then to investigate the detailed metabolic pathway with enzymes obtained from these bacteria. Results both with whole cells and partially purified enzymes have suggested the pathway of pyrimidine metabolism indicated in Fig. 1.

On the mechanism of pyrimidine metabolism by yeasts.

Knowledge of the mechanisms involved in pyrimidine metabolism is fundamental to the biochemistry of the nucleic acids. Cerecedo, Emerson, and Stekol (l-7) pioneered this field in their work with dogs. Their experiments, conducted by feeding the in vitro oxidation products of uracil and the estimation of excreted urea, culminated in the proposal of a mechanism for pyrimidine metabolism (8). More...

An ab initio quantum chemical investigation of TOMS nmr SHIELDING TENSORS IN Adenine-thymine, Adenine-uracil, Guanine-Cytosine & uracil-quartet: comparison between theoretical and experimental results

We have evaluated the NMR shielding tensors for A:T,G:C,A:U in Watson-crick, and U-quartet. We have computed NMR shielding tensors at B31YP level by using 6-31G(d) basis set. We have compute anisotropy and asymmetry in A:T,G:C,A:U and U-quartet. The NMR shielding tensors were calculated using the GIAO method. The natural bonding orbital analysis (NBO) were performed. NBO calculation have been ...

Metabolism of pyrimidine bases and nucleosides in the coryneform bacteria Brevibacterium ammoniagenes and Micrococcus luteus.

The metabolism of exogenous pyrimidine bases and nucleosides was investigated in Brevibacterium ammoniagenes and Micrococcus luteus with fluorinated analogs and radioactive precursors. Salvage of thymine and thymidine was found in M. luteus, but not in B. ammoniagenes. Exogenous uracil or uracil nucleosides, but not cytosine or cytosine nucleosides, were nucleic acid precursors for both bacteri...

Formation of cytosine glycol and 5,6-dihydroxycytosine in deoxyribonucleic acid on treatment with osmium tetroxide.

OsO4 selectively forms thymine glycol lesions in DNA. In the past, OsO4-treated DNA has been used as a substrate in studies of DNA repair utilizing base-excision repair enzymes such as DNA glycosylases. There is, however, no information available on the chemical identity of other OsO4-induced base lesions in DNA. A complete knowledge of such DNA lesions may be of importance for repair studies. ...

Life without dUTPase

Fine-tuned regulation of the cellular nucleotide pools is indispensable for faithful replication of Deoxyribonucleic Acid (DNA). The genetic information is also safeguarded by DNA damage recognition and repair processes. Uracil is one of the most frequently occurring erroneous bases in DNA; it can arise from cytosine deamination or thymine-replacing incorporation. Two enzyme activities are prim...
